When I did my student teaching, the 100th day project was for the first graders to collect 100 of something- and then use their collection to create something else.
A few that I remember were a snowman made from 100 cotton balls, a caterpillar made from 100 pom-pom balls, a building made from 100 legos, and 100 Fruit Loops glued to a poster in the shape of a flower.
I want to assign this project again, but the group of parents I have this year are not very motivated. I'm trying to compile a list of ideas (that I'll share after giving them time to brainstorm on their own) and I'm hoping some of the creative teachers here will help me add to the list!!

We had our first graders each bring in a 100 of an item. The note home specifically said to make sure the item was inexpensive. We practiced counting, sorting and patterning. When we were finished each student handed one of their items to every other student. After that, I gave them a giant piece of paper and they made 100 day artwork out of the items. The kids loved it.

I made sure to have a box of Cocoa Puffs on hand. I gave the students that didn't bring anything a 100 Cocoa Puffs.
